Discussions about the achievements and limitations of the various approaches to the development of intelligent systems can have an essential impact on empirically based research, and with that also on the future development of computer technologies. However, such discussions are often based on vague concepts and assumptions. In this context, we claim that the proposed `three-world ontology' offers the most appropriate conceptual framework in which the basic problems concerned with cognition and computation can be suitably expressed and discussed, although the solutions of some of these problems seem to lie beyond the horizon of our current understanding. We stress the necessity to differentiate between authentic and functional cognitive abilities; although computation is not a plausible way towards authentic intelligence, we claim that computational systems do offer virtually unlimited possibilities to replicate and surpass human cognitive abilities on the functional level.  相似文献

The toxic effects of photoproducts formed upon the photolysis of 2- and 4-chlorophenol (CP) frozen solutions in polycrystalline ice phase were determined with a bacterial luminescence test (Vibrio fisheri), and in vitro biomarker assay for dioxin-like effects (inductions of AhR-dependent luciferase in H4IIE-luc cells) and compared to the toxic effects of products of the same photoreaction in aquatic phase. Coupling photoproducts formed in ice samples (3'-chlorobiphenyl-2,4'-diol and 3-chlorobiphenyl-2,2'-diol from 2-CP photolysis and 5-chlorobiphenyl-2,4'-diol from 4-CP photolysis) were found to be more toxic to V. fisheri than parent CPs and elicited significant inductions of dioxin-like effects (the effective concentrations EC50 approximately 3 x 10(-5) mol L(-1) corresponded to known weaker ligands of AhR, such as nonplanar polychlorinated biphenyls or polycyclic aromatic hydrocarbons). To complete the picture, a photoproduct formed from 4-CP (5-chlorobiphenyl-2,4'-diol) was synthesized, and a detailed toxicity assessment with purified compound confirmed the results obtained with irradiated samples. Our findings support a recently proposed model according to which solar radiation can trigger the formation of new types of organic pollutants in polar ice or tropospheric ice cloud particles, presenting possibly greater risk to the environment than the parent compounds.  相似文献

The aim of this study was to find out whether very low doses of nephrotoxic and hepatotoxic mycotoxins ochratoxin A (OTA) and fumonisin B1 (FB1) induce oxidative stress in rat kidney and liver and whether their effect is synergistic. Rats were treated orally with OTA (5 ng/kg b.w. and 50 microg/kg b.w.) and FB1 (200 ng/kg b.w. and 50 microg/kg b.w.), or their combinations. Malondialdehyde (MDA) and protein carbonyls (PCs) concentration in kidney was affected with lower dose of OTA than in liver (p<0.05). FB1 did not affect MDA and PCs concentrations in the liver, while in the kidney both FB1 doses increased MDA concentration (p<0.05). The combination of the lower doses of OTA+FB1 increased the MDA and PCs concentration both in the liver and the kidney, compared to controls and animals treated with respective doses of mycotoxins (p<0.05). The combinations of mycotoxins reduced the catalase activity only in the kidney when compared to controls (p<0.05). In contrast to the increased kidney concentrations of MDA and PCs even with very low doses of OTA and FB1, the activity of catalase and SOD does not change. Combinations of OTA+FB1 affected almost all parameters, which indicates their potential to produce oxidative damage.  相似文献

A mathematical model is developed to describe the globular transfer in gas metal arc welding (GMAW). This work is both theoretical and experimental. Using the volume-of-fluid (VOF) method, the fluid-flow and heat-transfer phenomena are dynamically studied during the following processes: droplet formation and detachment, impingement of a droplet on a solid substrate, impingement of multiple droplets on the molten pool, and solidification after the arc extinguishes. A He-Ne laser, in conjunction with the shadow graphing technique, is used to observe the metal transfer processes. Theoretical predictions and experimental results are in close agreement, suggesting that the theoretical treatment of the model is good.  相似文献

A new method for dealing with the effects of quantization in a subband system is proposed. It uses the "gain plus additive noise" linear model for the Lloyd-Max quantizer. Based on this, it is demonstrated how, by an appropriate choice of synthesis filters, one can cancel all signal-dependent errors at the output of the system. The only remaining error is random in nature and not correlated with the input signal. We therefore have a tradeoff between the error being only random or having signal-dependent components as well (since the error variances in both cases are comparable). As a result of having only a random error, it is possible to reduce this error using, for example, a noise removal technique. The result is then extended to the case where the input is a multidimensional signal, and arbitrary sampling lattices are used, as well as to the QMF (alias cancellation) case. To demonstrate the validity of the proposed approach, two types of experiments on images are carried out: In a toy example, it is shown that using noise removal could be beneficial. For a more realistic coding scheme, however, it is demonstrated that even in the case when the model is no longer valid (when some of the subbands are discarded), the output error is still much less correlated with the input signal as opposed to the commonly used subband system, while visually, the reconstructed images look very similar.  相似文献

Biologically functional Ras isoforms undergo post-translational modifications starting with farnesylation of the most C-terminal cysteine. Combined with further processing steps, this isoprenylation allows for the anchoring of these proteins in endomembranes, where signal transduction events take place. The specific localization is subject to dynamic regulation and assumed to modulate the activity of Ras proteins by governing their spatiotemporal distribution. The delta subunit of phosphodiesterase (PDEdelta) has attracted attention as a solubilization factor of isoprenylated Ras. In this study, we demonstrate that critical residues in the putative isoprenoid pocket of PDEdelta can be mapped by coupling with a semisynthetic N-Ras lipoprotein in which the native farnesyl group of the processed protein was replaced by a photoactivatable geranyl benzophenone moiety. The crosslinked product included parts of beta-sheet 9 of PDEdelta, which contains the highly conserved amino acids V145 and L147. Modeling of the PDEdelta-geranyl benzophenone (GerBP) complex supports the conclusion that the photolabeled sequence is embedded in the putative isoprenoid pocket of PDEdelta.  相似文献

Hearing loss impacts a large proportion of the global population. Damage to the inner ear, in particular the sensitive hair cells, can impact individuals for the rest of their lives. There are very limited options for interventions after damage to these cells has occurred. Targeted gene delivery may provide an effective means to trigger appropriate differentiation of progenitor cells for effective replacement of these sensitive hair cells. There are several hurdles that need to be overcome to effectively deliver these genes. Nanoencapsulation technology has previously been used for the delivery of pharmaceuticals, proteins and nucleic acids, and may provide an effective means of delivering genes to trigger appropriate differentiation. This review investigates the background of hearing loss, current advancements and pitfalls of gene delivery, and how nanoencapsulation may be useful.  相似文献

A closed-loop system is developed to control the weld fusion, which Is specified by the top-side and back-side bead widths of the weld pool. Because in many applications only a top-side sensor is allowed, which is attached to and moves with the welding torch, an image processing algorithm and neurofuzzy model have been incorporated to measure and estimate the top-side and back-side bead widths based on an advanced top-side vision sensor. The welding current and speed are selected as the control variables. It is found that the correlation between any output and input depends on the value of another input. This cross coupling implies that a nonlinearity exists in the process being controlled. A neurofuzzy model is used to model this nonlinear dynamic process. Based on the dynamic fuzzy model, a predictive control system has been developed to control the welding process. Multipass dynamic MRI and pharmacokinetic modeling are used to estimate perfusion parameters of leaky capillaries. Curve fitting and nonblind deconvolution are the established methods to derive the perfusion estimates from the observed arterial input function (AIF) and tissue tracer concentration function. These nonblind methods are sensitive to errors in the AIF, measured in some nearby artery or estimated by multichannel blind deconvolution. Here, a single-channel blind deconvolution algorithm is presented, which only uses a single tissue tracer concentration function to estimate the corresponding AIF and tissue impulse response function. That way, many errors affecting these functions are reduced. The validity of the algorithm is supported by simulations and tests on real data from mouse. The corresponding nonblind and multichannel methods are also presented.  相似文献

In this investigation, hybrid laser/arc welding (HLAW) was employed to join 8-mm-thick high-strength quenched and tempered steel (HSQTS) plates in the butt-and T-joint configurations. The influences of welding parameters, such as laser power, welding speed, stand-off distance (SD) between the arc of gas metal arc welding, and the laser heat source on the weld quality and mechanical properties of joints, were studied to obtain non-porous and crack-free fully-penetrated welds. The weld microstructure, crosssection, and mechanical properties were evaluated by an optical microscope, and microhardness and tensile tests. In addition, a finite element model was developed to investigate the thermal history and molten pool geometry of the HLAW process to join the HSQTS. The numerical study demonstrated that the SD had a paramount role in good synergy between the heat sources and the stability of the keyhole. For the butt-joint configuration, the results showed that, at a higher welding speed (35 mm/s) and optimum SD between the arc and laser, a fully-penetrated sound weld could be achieved. A non-porous weld in the T-joint configuration was obtained at a lower welding speed (10 mm/s). Microstructural evaluations indicated that the formation of residual austenite and the continuous network of martensitic structure along the grain boundary through the heat affected zone were the primary reasons of the softening behavior of this area.
